  • The medium defined by Kao and Michayluk was designed to grow cells and protoplasts of Vicia hajastana at a very low population -density in liquid media.
  • The inability of the plant cells to grow at a very low population density may be caused by excessive diffusion of metabolic intermediates into the medium, resulting in their dilution in the cell to a level below that required for survival.
  • Vicia cells were able to grow at an initial population density of 1-2 cells/ml when the mineral salt medium was enriched with organic acids, sugars, sugar alcohols, growth regulators, amino acids and other organic compounds.
  • The percentage of cell division could be increased by raising the -concentration of CaCl2 from 1 mM, as in Gamborg B5, to 5 mM.
  • Calcium may play an important role in the proces of cell division because of its ability to preserve the structural and functional integrity of plant cell membranes.
